QK-全科广谱治疗仪对冠心病病人血液过氧化反应及红细胞变形性的影响

提要 目的和方法:本实验检测了35例冠心病病人使用QK-全科广谱治疗仪治疗前后血中红细胞超氧化物歧化酶(SOD)活力;全血谷胱苷肽过氧化物酶(GSH-Px)、过氧化氢酶(CAT)活力;血浆总抗氧化能力(AOA);红细胞膜丙二醛(MDA)含量;红细胞滤过指数(IF)的变化。结果:治疗前全血GSH-Px、CAT活力、血浆AOA明显下降,红细胞膜MDA含量明显升高,红细胞IF明显升高。治疗后与治疗前比较,全血GSH-Px、CAT活力及血浆AOA明显升高,红细胞MDA含量明显降低,红细胞IF明显降低。结论:使用QK-全科广谱治疗仪治疗能减轻病人血液过氧化反应,改善红细胞变形性(ED)。
Blood peroxide reaction and erythrocyte deformability before and after the management with qK-wind band spectrum medicoapparatus in CHD patients

Abstract In order to study the effect of QK-wind band spectrum medicoapparatus (QK-WBSM) on the blood peroxydation and erythrocyte defomability (ED) in coronary heart disease (CHD) patients.the superoxide dismutase (SOD) ability of erythrocyte,glutathione peroxidase(GSH-Px),cataiase (CAT) activity of blood ,antioxidite activity (AOA) of plasma,molondialehyde (MDA) concent of erythrocyte membrane and erythrocyte filtration (IF) were measured in the SOD activity of erythrocyte,patients with CHD before and after treatment using QK-WBSM.The results showed that in the patients the GSH-Px,CAT activity of blood and AOA of plasma were much lower than those of the control (P<0.05),the MDA concents of erythrocyte membrane and erythrocyte IF were much higher than those of control (P<0.01).After treatment with QK-WBSM the SOD activity of erythrocyte,GSH-Px,CAT activity of blood and AOA of plasma significantly higher than those of the group before treatment (P<0.05),the MDA concents of erythrocyte membraneand and erythrocyte IF were much lower than those of the group before treatment (P<0.05,P<0.01).The results indicated that QK-WBSM can reduce the peroxide reaction of blood and enchance ED in the CHD patients.

研究资料表明,冠心病(coronary heart disease,CHD)病人血液中过氧化脂质含量升高,相关抗氧化酶类物质含量降低或活力下降[1~3]。心肌缺血时红细胞变形性(Erythrocyte defomability,ED)明显下降,ED下降是加重心肌缺血性损伤的重要因素[4,5]。临床资料表明,QK-全科广谱治疗仪(QK-wind band spectrum medicoapparatus)具有明显的调节人体生理功能作用,我们检测了35例CDH心绞痛病人,使用QK-全科广谱治疗仪治疗前后红细胞超氧化物歧化酶(Superoxide disutase,SOD)、全血谷胱苷肽过氧化物酶(Glutathione peroxidase,GSH-Px)、过氧化氢酶(Cataiase,CAT)活力、血浆总抗氧化能力(Antioxidite ability,AOA)、红细胞膜丙二醛(Molondialehyde,MDA)含量、红细胞滤过指数(Filtration,IF)的变化,以期探讨QK-全科广谱治疗仪对冠心病病人的治疗意义。
1 资料与方法
1.1 临床资料
根据WHO诊断标准选择CHD病人35例,男18例,女17例,平均年龄(46.62±5.02)岁。所有病例于治疗前及治疗1月后晨起空腹采肘静脉血。对照组选择无慢性心脑血管疾病和内分泌、肾脏病史,血压、血脂及心电图均正常的志愿者23例,男14例,女9例,平均年龄(44.89±6.18)岁,晨起空腹采血。
1.2 方法
红细胞膜MDA含量测定:采用化学比色法。红细胞SOD活力测定:采用化学比色法。全血CAT活力测定:采用化学比色法。全血GPD测定:采用化学比色法。血浆AOA测定:采用化学比色法。以上检测所有试剂为南京建成生物研究所提供的试剂盒,并按说明书方法进行测定。红细胞IF测定:按戴稼禾等[6]方法检测,作为红细胞变形性(ED)的指标,IF愈大则ED愈小,反之IF愈小ED愈大。统计学处理:实验数据以±s表示,采用t检验及相关分析进行差异显著性检验。
2 结果
红细胞膜MDA含量的变化:治疗前病人血液红细胞MDA含量较对照组明显增高(P<0.01),治疗后较治疗前显著降低(P<0.01),见附表。血液抗氧化物质的变化:治疗前与对照组比较及治疗前后红细胞SOD活力未见明显改变(P>0.05)。治疗前全血GPD、CAT活力、血浆AOA较对照组明显降低(P<0.01),治疗后较治疗前明显提高(P<0.05),见附表。红细胞变形性的变化:治疗前红细胞IF较对照组明显增高(P<0.01),治疗后较治疗前明显降低(P<0.05),见附表。红细胞IF与红细胞膜MDA含量呈明显正相关(r=0.687,P<0.01)。

3 讨论
ED作为从血液流变学角度反映疾病发生发展及预后的一项重要指标愈来愈受到人们的重视。研究资料表明,ED降低可发生小血管阻塞,增大临界管径的数值,通过逆转现象使冠脉阻力增大,加剧心肌缺血性损伤[5]。也是心肌损伤面积扩大和预后不良的重要原因之一[4]。CHD病人由于酶类及非酶类抗氧化物质水平的降低,过氧化反应加剧,MDA含量增高,导致其血管内皮损伤,脂质在动脉壁沉积,血小板于损伤处聚集并释放生长因子,刺激平滑肌细胞增生,MDA尚可通过抑制PGI的合成,使PGI与TXA失衡,促发动脉粥样硬化[7,8]。同时,MDA还可与红细胞的含有游离氨基酸的蛋白质、磷脂酰乙醇胺等形成Schiff硷,使分子间发生交联,增加膜的钢性,降低膜的流动性,还可使蛋白质变性,酶失活,膜通透性发生改变,膜结构及功能受损而降低ED[9~12],红细胞变形能力下降,且易发生聚集,促进微循环障碍,加剧心肌缺血性损伤。本文结果表明,CHD病人IF明显升高,且IF升高与红细胞膜MDA增高呈显著性正相关,说明ED的下降与血液抗氧化能力降低及其所致的红细胞膜氧化损伤有关。故寻找提高血液抗氧化能力,减轻红细胞膜氧化损伤改善ED的有效措施对改善微循环灌注,减轻心肌缺血性损伤,缩小损伤面积,改善预后有重要意义。采用QK-全科广谱治疗仪治疗可使CHD病人红细胞膜MDA含量较治疗前明显下降:全血GSH-Px、CAT活力较治疗前明显增高、血浆AOA较治疗前明显升高;红细胞IF较治疗前明显下降。说明病人血液抗氧化水平提高,红细胞膜氧化损伤程度减轻,ED明显改善。可见QK-全科广谱治疗仪对CHD病人的治疗有一定的实际应用价值。
